The novel Requiem for a Dream written in 1978 by Hubert Selby Jr. was the source used by Darren Aronofsky for the film adaption made in 2000.
Marion did not become a full-time prostitute, but had sex with Big Tim to get the drugs to satisfy her addiction. We see Marion sell herself three times throughout the movie, once with her shrink, once with Big Tim, and finally in the ass-to-ass scene at Big Tim's party.Note: The ass-to-ass scene is only in the Director's Cut.
At the end Harry had to get his arm amputated because of an infection from his heroin addiction. He got gangrene from injecting heroin straight into the wound on his arm, and the infection spread. - Harry did not rotate his injection sights so rather then him getting gangrene he would have developed an abscess and he was still digging (injecting) into that abscess and not seeking the correct medical attention for it, it got infected leading it to spead the infection throughout that arm needing it to be amputated.
